Analytical Instrumentation - Spectrofluorimeter

In the following video,we continue from the previous video where we learnt about the basic principle of fluorescence and phosphorescence. The generation of fluorescent and phosphorescent emissions is discussed in detailed using an energy level diagram of a molecule. The fluorescence and phosphorescence emitted by a sample under test is used to perform qualitative and quantitative analysis on the analyte. It is performed using spectrofluorimeter. The basic principle of measurement of fluorescence is also explained in the previous video.
In the current video, we understand how a spectrofluorimeter is constructed. The different components used to construct the device starting from light sources like xenon lamps, mercury vapour lamps, lasers and LED's , the monochromators like prism monochromators or diffraction monochromators are used. The sample is excited using a light source to generate fluorescence. The generated fluorescence is measured using a detector like a PMT (Photomultiplier tube).
After the construction, the working of the spectrofluorimeter is explained with the help of an animated photon of light.
