The first and a very basic instrument called single beam photometer or colorimeter is explained in the following animated video. The photometer is used to determine the concentration of the unknown sample using the principle of absorption of light by the sample under test. The instrument is called photometer because it uses filters as opposed to monochromators to isolate the wavelenghts of light falling on the sample or analyte. The light gets absorbed by the sample and remaining light is transmitted which falls on the detector. The detector can be photovoltaic or photoemissive cell. Using the reference sample with known concentration and readings from the photocell, Beer-Lambert's law is used to determine the concentration of the unknown analyte.
